Depending where you are staying in Phuket your ladyboy access may vary. However most of the ladyboys in Phuket are in Patong which is the main city where tourists may stay. In Patong most of the ladyboys are going to be found around the Bangla road area. Unfortunately I do not think there is any dedicated ladyboy bars in Phuket anymore but there is many bars in the Bangla area that have some ladyboys on staff. So if you like bar hopping then you should be able to find some ladyboys. There is also usually many on the street on Bangla road but be careful of freelancers from the street. If you only find one ladyboy you like you can ask them if they have a friend.
Watch for bars that have ladyboy cabaret shows and those may be a good option.
Soi Paradise is kind of a gay area but they often have ladyboy cabaret shows also.
Like in Bangkok can try to meet them online on sites such as Thai Friendly but then you can't be sure what you are getting and it may be harder to set up a threesome.
Again f you find one ladyboy you can ask if they have a friend to join, this way you know they like each other.
There is a couple other options in Patong.
Tootsies ladyboys massage appears to still be open. Not sure how many ladyboys they have on staff these days.

All I can say is that Bangla road in Patong is the best place to find ladyboys. I haven't been there since Cocktails and Dreams bar closed which was the last dedicated ladyboy bar.
Hopefully somone else has more current information on Patong.
Bangla road and the streets off of it are the main place you will find ladyboys in Patong and Phuket.
Soi Seadragon and the side allies off Bangla road are good options to explore. Likely all bars with ladyboy will also have ladies and you should be able to spot them. Some bars may have many. Very likely to find a threesome with a lady and a ladyboy also.
Watch for any bars offering ladyboy cabaret shows as those would be good places to try.
Most Discos don't allow ladyboys but the Tiger Disco on the second floor allows ladyboys inside but they will be freelancers.
There will literally be ladyboys everywhere around the Bangla area and there is tons on the street but be careful there is high crime in the area. Many of the pick pocketers and scammers have turned out to be ladyboys working those streets. This is why find them in a bar can be safer than off the street.
However around Bangla there is lots of great ones on the street you just have to be careful.
